چکیده

In brain tumors, delivering nanoparticles across the blood-tumor barrier presents major hurdles. A clinically relevant MRI contrast agent, GdDOTA and a near-infrared (NIR) fluorescent dye, DL680 were conjugated to a G5 PAMAM dendrimer, thus producing a dual-mode MRI and NIR imaging agent. Systemic delivery of the subsequent nano-sized agent demonstrated glioma-specific accumulation, probably due to the enhanced permeability and retention effect. In vivo MRI detected the agent in glioma tissue, but not in normal contralateral tissue; this observation was validated with in vivo and ex vivo fluorescence imaging. A biodistribution study showed the agent to have accumulated in the glioma tumor and the liver, the latter being the excretion path for a G5 dendrimer-based agent.

Imaging brain tumor by dendrimer-based optical/paramagnetic nanoprobe across the blood-brain barrier.

A multimodal optical/paramagnetic nanoprobe, Den-Angio, was developed and demonstrated a capability to circumvent the blood brain barrier (BBB) and visualize brain tumors with high sensitivity in vivo. Den-Angio holds promise to pre-operatively localize brain tumors and make image-guided tumor resection possible during surgery.

A Prodrug-type, MMP-2-targeting Nanoprobe for Tumor Detection and Imaging

Tumor-associated proteases (TAPs) have been intensively studied because of their critical roles in cancer development. As a case in point, expression of matrix metalloproteases (MMP) is significantly up-regulated in tumorigenesis, invasion, and metastasis among a majority of cancers. Here we present a prodrug-type, MMP-2-responsive nanoprobe system with high efficiency and low toxicity for dete...

Magnetic Graphene Oxide Nanocarrier as a drug delivery vehicle for MRI monitored magnetic targeting of rat brain tumors

Introduction: Glioblastoma multiform is the most common malignant brain tumor, with an invasive nature. Despite the development of conventional therapies such as surgery, radiotherapy and chemotherapy, because of high recurrence rates, the prognosis remains very poor. Over the last decade, nanotechnology has represented an innovative method as nanoparticle-based drug delivery ...

Y1 receptor ligand-based nanomicelle as a novel nanoprobe for glioma-targeted imaging and therapy.

Due to the molecular and cellular heterogeneity of glioma, discovery of novel targeted sites and ligands for glioma imaging and therapy remains challenging. Neuropeptide Y (NPY) Y1 receptors (Y1Rs) are highly over expressed in various brain tumors including glioma, and can serve as potential targeting sites for glioma imaging and therapy. Here, we show by in vivo fluorescent imaging that a high...

Specific targeting of brain tumors with an optical/magnetic resonance imaging nanoprobe across the blood-brain barrier.

Nanoparticle-based platforms have drawn considerable attention for their potential effect on oncology and other biomedical fields. However, their in vivo application is challenged by insufficient accumulation and retention within tumors due to limited specificity to the target, and an inability to traverse biological barriers. Here, we present a nanoprobe that shows an ability to cross the bloo...
